Counts one fragment per transcript from the 3′ end, so reads map directly to expression level and far less sequencing depth is needed than whole-transcriptome library prep. It handles low-input, degraded and blood samples that defeat full-length methods, and remains the most cost-efficient route to differential expression across large sample sets.
